For satellite ocean color to be reliable and used in these and other important environmental applications, the data must be trustworthy and high quality. Pre-flight and on-board calibration of satellite ocean color sensors is conducted; however, once in orbit, the data quality can only be fully assessed via independent calibration and validation activities using surface measurements. These measurements therefore need to be at least as high quality as the satellite data, which necessitates SI traceability and a full uncertainty budget. This is the basis for fiducial reference measurements (FRMs) and the FRM4SOC project, which was an European Space Agency (ESA) initiative to establish and maintain SI-traceable ground-based FRM for satellite ocean color, thus providing a fundamental contribution to the European system for monitoring the Earth (Copernicus). Overall, this is primarily due to legislative efforts to improve the quality of water bodies and oceans. Remote sensing has played a key role in the development of such approaches—from the use of drones for vegetation mapping to autonomous vessels for water quality monitoring. Within the specific context of vegetation characterization, the wide range of available observations—from satellite imagery to high-resolution drone aerial imagery—has enabled the development of monitoring and mapping strategies at multiple scales (e.g., micro- and mesoscales). However, our understanding of the microbiology of this system, especially the subsurface “deep biosphere” component of it, has traditionally been limited by sample availability and quality. Title A name given to the resource Chapter 14 Coupled Enzyme Activity and Thermal Shift Screening of the Maybridge Rule of 3 Fragment Library Against Trypanosoma brucei Choline Kinase; A Genetically Validated Drug Target Subject The topic of the resource pharmacology Description An account of the resource In this study we interrogate 630 compounds of the Maybridge Rule of 3 Fragment Library for compounds that interact with, and inhibit TbCK. The Maybridge Rule of 3 Fragment Library is a small collection of quantifiable diverse, pharmacophoric rich, chemical entities that comply with the following criteria; MW ≤ 300, cLogP ≤ 3, H-Bond Acceptors ≤ 3, H-Bond Donors ≤ 3, Rotatable bonds (Flexibility Index) ≤ 3, Polar Surface Area ≤ 60 Å2 and aqueous solubility ≥ 1 mM using LogS and high purity (≥ 95%). Comparisons between two different screening methods, a coupled enzyme activity assay and differential scanning fluorimetry, has allowed identification of compounds that interact and inhibit the T. brucei choline kinase, several of which possess selective trypanocidal activity. Screening of a comparatively small fragment library by two different screening methods has allowed identification of several compounds that interact with and inhibit TbCK, a genetically validated drug target against African sleeping sickness. Some of the inhibitory fragments were also selectively trypanocidal, considering these are relatively simple molecules with no optimization, finding low μΜ inhibitors is very encouraging. Moreover some of the morphological phenotypes of these trypanocidal compounds include cell-cycle arrests similar to those observed for the TbCK conditional knockout grown under permissive conditions. Title A name given to the resource Chapter 3 Vitamin C Alimentation via SLC Solute Carriers Subject The topic of the resource Pharmacology Description An account of the resource Although mortality rates from pneumonia and sepsis are decreasing in many parts of the world, the incidence of these conditions continues to rise, likely due to an increasingly ageing population. Case reports and observational studies indicate a strong association between vitamin C status and infectious conditions, with vitamin C deficiency predisposing individuals to infections, and conversely, infections precipitating a diminished vitamin C status. The requirements for vitamin C increase with the severity of the infectious condition, with gram amounts required to normalize the vitamin C status of critically ill patients. Over the past few years, there has been renewed interest in the role that vitamin C may play in severe infectious conditions. Vitamin C has a plethora of physiological effects, primarily due to its activity as a cofactor for a growing group of biosynthetic and regulatory enzymes, with the potential to regulate thousands of genes and cell signaling pathways. Small intervention studies have indicated that administration of gram doses of vitamin C to patients with pneumonia and sepsis improves organ function and potentially decreases mortality, although this remains to be confirmed in larger trials. The long-term quality of life outcomes of these patients also remains to be determined.
